Neospora caninum truncated recombinant proteins formulated with liposomes and CpG-ODNs triggered a humoral immune response in cattle after immunisation and challenge

چکیده

Abortions caused by Neospora caninum are a serious problem in cattle production and require effective immunoprophylaxis. The objective of this work was to assess the humoral immune response four recombinant (r) N. antigens after immunisation challenge. MIC1 MIC3 proteins from micronemes, SRS2 surface tachyzoites, GRA7 dense granules were expressed as truncated Escherichia coli. Cationic liposomes (Lip) CpG oligodeoxynucleotides (CpG-ODNs) used adjuvant. Steers assigned three groups six steers each inoculated twice subcutaneously, 21 days apart. rP + Lip CpG-ODN group received rMIC1, rMIC3, rSRS2 rGRA7 formulated with adjuvant; adjuvant alone; PBS sterile phosphate-buffered saline. All subcutaneously challenged NC-1 strain 35 second dose immunisation. developed specific IgG, IgG1 IgG2 against After challenge, IgG rMIC1 rMIC3 detected all groups. remained high challenge group. Indirect ELISA anti-N. antibodies groups, highest level being vaccine using immunogenic enhanced vaccinated cattle.

Serological response over time to recombinant Neospora caninum antigens in cattle after a neosporosis-induced abortion.

Recombinant Neospora caninum tachyzoite antigens were evaluated in an enzyme-linked immunosorbent assay (ELISA) for recognition by serum antibodies (Ab) from Neospora-infected cattle. Serum samples were obtained every 2 to 3 weeks for 8 to 15 months from 10 cows with histories of Neospora-associated abortion. Serum samples were also obtained from offspring of these animals and from a large numb...

Development of a diagnostic method for neosporosis in cattle using recombinant Neospora caninum proteins

BACKGROUND Neosporosis is an infectious disease primarily of cattle and dogs, caused by intracellular parasite, Neospora caninum. Neosporosis appears to be a major cause of abortion in dairy cattle worldwide and causes to huge economic loss to dairy industry. RESULTS Recombinant surface associated antigen 1 (NcSAG1), NcSAG1 related sequence 2 (NcSRS2) and the dense granule antigen 2 (NcGRA2) ...

Humoral immune response in pregnant heifers inoculated with Neospora caninum tachyzoites by conjunctival route.

The aim of this study was to compare systemic humoral immune responses in pregnant heifers inoculated with Neospora caninum tachyzoites by conjunctival and intravenous routes. Twenty nine heifers separated in three experimental groups were studied: Group 1 (n=10 animals) and Group 2 (n=9 animals) were inoculated with 10(8) of N. caninum tachyzoites by conjunctival and intravenous routes at 5th ...

Development of maternal and foetal immune responses in cattle following experimental challenge with Neospora caninum at day 210 of gestation

This study examined the immunological responses of pregnant cattle and their foetuses following an experimental challenge with live Neospora caninum tachyzoites at day 210 of gestation. Animals were bled prior to and weekly throughout the experiment and sacrificed at 14, 28, 42 and 56 days post inoculation (dpi). At post mortem examination, samples of lymph nodes and spleen were collected from ...

Maternal and foetal immune responses of cattle following an experimental challenge with Neospora caninum at day 70 of gestation

The immune responses of pregnant cattle and their foetuses were examined following inoculation on day 70 of gestation either intravenously (iv) (group 1) or subcutaneously (sc) (group 2) with live NC1 strain tachyzoites or with Vero cells (control) (group 3). Peripheral blood mononuclear cell (PBMC) responses to Neospora antigen and foetal viability were assessed throughout the experiment. Two ...
